Abstract : 5-Fluorouracil (5-FU) has been introduced as the most common anticancer therapy despite that its response rate as a single agent is usually less than 20%. Lactobacillus (L) rhamnosus bacteria exhibit a great impact on the gastrointestinal tract infections and cancer prevention. This study illustrates the histological and Immunological changes associated with the treatment of L. rhamnosus combined to 5-FU on mice colon cancer. Fifty male mice were classify into 5 groups as follow; (A group) mice injected with Azoxymethane (AOM) with a weekly dose of 10mg/kg for four weeks to induce colon cancer, (AL group) mice injected with AOM and orally given L. rhamnosus alone, (AF group) mice injected with AOM and injected 5-FU, (AFL group) mice injected with AOM and treated with both L. rhamnosus and 5-FU, and finally (C group) untreated control mice. A reduction in the inflammation features and a normal histological structure was observed in the colon of AFL mice group compared with other treated mice groups, which seemed more similar to the untreated mice group. A significant downregulation in the level of Kras and T-Regulatory cell /IL10 transcriptions, associated with an improvement in the innate and the adaptive immune responses through increasing in the TLR2 and Th1/IFNγ transcription was verified in the intestine mucosa of AFL mice group. TNFα and IL6 elevated significantly in the serum of AFL mice groups compared to A and AF mice groups. This study provide evidence about the high potential immunological influence of L. rhamnosus during the usage of 5-FU as anticancer drug and illustrate the positive impact on colon cancer suppression.
