Abstract : The list of indications for the clinical use of botulinum toxin (BOTOX) has expanded widely. Botox injections are used to control a large number of muscle disorders characterized by abnormal involuntary spasmodic contractions, e.g. focal dystonia and dysfunction of extra-ocular muscles, in addition to correction of skin wrinkles and treatment of excessive sweating. As the toxin-induced cure is temporary, patients need to receive more than one injection at timely intervals. In view of such extensive use of botulinum toxin, it becomes crucially important to have a comprehensive understanding of the sequence of events when this toxin is injected into muscle. In another respect, Botox injections are now being used for alleviating spasticity associated with cerebral palsy in children. This has raised the question whether Botox injections in juvenile muscle produce the same structural changes as in adult muscles! And if this was the case, do these alterations follow the same time scale? In the present study, electron microscopy and immuno-histochemical techniques were used to investigate and contrast the structural changes induced by Botox injections in juvenile versus adult rat skeletal muscle. The results indicate that the same set of structuctural changes took place in both cases following Botox injection. However, the time scale of these changes was found to be significantly different in juvenile versus adult muscle.
